Principle The Test is a membrane based sandwich immunochromatographical assay. An anti-CDV capture antibody is immobilized on the membrane in the test zone. A gold-labeled anti-CDV antibody is imbedded in a sample pad where the sample is applied. After sample application, the gold labeled antibody binds to the antigen in the sample to form an antigen-antibody complex. As the sample-gold labeled antibody mixture migrates along the membrane, the antigen-labeled antibody complex is captured by the immobilized anti-CDV antibody in the test zone to form an antibody-antigen-gold labeled antibody sandwich as indicated by the appearance of a magenta test line. The absence of the test line indicates a negative test. In the control zones of the membrane, a magenta control line appears in every valid test indicating that the test is properly performed and reagents are functional as specified. Intended Use Canine Distemper (CDV) Test is designed to detect the antigens of CDV in canine ocular or nasal discharge. a. Allow test card, reagent, specimen and controls to equilibrate to room temperature (15-30°C) before testing. b. Place the test card on a clean and level surface. c. Obtain the treated specimen by using a disposable pipette, and transfer 4 drop (100 µl) of specimen into the sample/reagent port of the test card. Test results can be inspected in 2 methods : A. Visual: Visual inspect by eye. B. Reader: Analyze with veterinary test reader. A. Visual Visual inspect the test results within 10 mins. Contents (10 tests/kit) Individually foil pouched test devices: 10 units Test buffer tube (0.4ml/tube): 10 units Disposable droppers: 10 units Swabs for sample collection: 10 units Instructions for use Control Line (C): The line should always appear. If this line does not appear, the test should be considered invalid. This is probably because of impure buffer and the lack of specimen. It should be tested again with new buffer. 1) Specimen Collection and Preparation Use a new swab to collect canine tears (or eye mucus), or snivel. Insert the applicator with the specimen into the collection tube containing 0.4mL of buffer. Negative: Control line only appears. ! Positive: Both Test and Control lines appear. ! Swirl the buffer with the swab for 10 seconds to ensure that the specimen is completely released. Invalid Results requiring Retesting a) Both Test and Control lines do not appear. ! If the treated specimen is not immediately tested, store the collection tube between 2ºC and 8ºC. Shake or vortex the mixture again before testing. b) Only Test Line Appears and Control line does not.
