BIORON life science Description: AntiTaq antibodies are used in various PCR methods to block the activity of Taq polymerase at ambient temperature, until the antibody is denaturated at temperatures above 60 °C. This allows convenient reaction set-up without additional cooling of the polymerases, which improves the specific amplification of DNA in the PCR. The monoclonal antibodies of type IgG are produced from a hybridoma clone with a high purity and are free of aggregates and endotoxins. Concentration: 2 mg/ml in storage buffer with 50 % glycerol Application: Antibody against Taq polymerase to block the enzyme activity at ambient temperature The amount of antibodies required to inhibit Taq polymerase activity depends on the amount of enzyme used and therefore can vary between different polymerases. The exact ratio can be found empirically: mix 1 unit of antibody with 1 unit of polymerase and check PCR performance with serial dilutions and a HotStart test. Repeat with different amounts of antibody and polymerase until the optimal ratio is found. Unit definition: One unit is defined as the amount of AntiTaq antibodies required to block 50 % activity of 1 ^g of Taq DNA Polymerase at 37 °C. 2300 Units of specific activity are equal to 1 mg of antibodies.
