Salt crystals may precipitate due to storage at 2 - 8 °C or freezing. Therefore, to prepare the working solution, the Stripping Buffer must be warmed to room temperature beforehand, which will dissolve any salts that may have precipitated. The buffer should be thoroughly mixed again by shaking immediately before use. To remove the antibodies, the blot membrane is placed in a dish or container with Stripping Buffer and incubated at room temperature for approx. 30 - 60 minutes with gentle shaking. After stripping the membrane has to be washed with a wash buffer (e.g. Washing Buffer PBS, article no. 140 or Washing Buffer TRIS, article no. 145) and can be used for a second detection. If you detect with alkaline phosphatase, the wash buffer should not contain phosphates, like Washing Buffer TRIS. Important: the incubation conditions mentioned are guide values that may have to be adjusted by the user. Incubation times depend on the properties of the antibodies used.
