Principle of Operation
The NucleoCounter® uses an integrated fluorescence microscope to detect signals from propidium iodide (PI) bound to DNA. The results represent the total cell concentration. The SCC-Cassette™ contains immobilized PI, which stains the cellular DNA when the cassette is loaded with the cell lysate. The stained mixture is transferred to the measurement chamber, where green light excites the PI-DNA intercalation, and the emitted red light is detected by a CCD camera to determine the cell count.

Sample Preparation and Analysis
  1. Prepare the sample by mixing equal amounts of milk and Reagent C (1:1 dilution).
  2. Load the SCC-Cassette™ with the diluted sample by immersing the tip and pressing the piston.
  3. Place the cassette in the instrument and press 'Run'. The somatic cell count is displayed and can be printed or transferred to a PC.

Sample Preparation The sample is easily prepared using equal amount of milk sample and Reagent C (1:1 dilution), followed by inversion of the sample preparation tube to assure mixing. Load the SCC-Cassette™ with the dilluted milk sample by immersing the tip of the cassette into the solution and pressing the piston. Place the SCC-Cassette™ in the instrument and press the “Run” key.
