Clean Plasmid TR DNA Kit

Clean Plasmid TR DNA Kit

Product catalog summary
Procedure: The process begins with resuspending and lysing bacteria using the CleanNA buffer system. After lysis, samples are centrifuged, and the supernatant is transferred to a new plate. Optionally, lysate clearance beads can be used to remove cell debris. Plasmid DNA binds to magnetic beads in the presence of a binding buffer, and the beads are separated using a magnetic device. After washing, plasmid DNA is eluted with an elution buffer.
Description: The Clean Plasmid TR DNA Kit utilizes a magnetic bead-based system to extract high-quality, endotoxin-free plasmid DNA. It is compatible with various liquid handling workstations, ensuring robustness and reproducibility. Plasmid yields depend on the E. coli strain, plasmid copy number, and growth conditions, with typical yields of 10-15 µg from 1 mL LB medium cultures. The DNA is suitable for transfections, Sanger sequencing, restriction enzyme digestion, and other molecular biology procedures.
Downstream Applications: The kit supports applications such as transfection, PCR, Sanger sequencing, and restriction digestion.
Ordering Information: The kit is available in two sizes: 96 preps (Catalog # CPLT-D0096) and 384 preps (Catalog # CPLT-D0384). Clearance beads are available in 5 mL (Part # CPLT-CB0005) and 50 mL (Part # CPLT-CB0050) sizes.
Plasmid DNA Yield: DNA was isolated from 0.8 mL cultures after 24 hours, with yields determined using a Denovix DS-11 spectrophotometer.
Endotoxin Levels: Endotoxin levels were measured using a Pierce LAL Chromogenic Endotoxin Quantitation Kit, with 50 µL of eluted DNA tested.
Comparative Analysis via Sanger Sequencing: DNA was isolated and sequenced to determine endotoxin levels, with sequencing reactions purified using CleanDTR and analyzed on an Applied Biosystems 3130XL instrument.
Comparative Performance: The Clean Plasmid TR DNA Kit showed superior performance in signal intensity and endotoxin levels compared to other methods, with an endotoxin level of 0.05 EU/µg, significantly lower than competitors.

Clean Plasmid TR DNA Kit-1

MAGNETIC BEAD BASED PLASMID DNA PURIFICATION SYSTEM Description The Clean Plasmid TR DNA Kit is based upon our proprietary magnetic beads based system to extract high quality and endo toxin free plasmid DNA. The system can easily be automated on many liquid handling workstations (e.g. Beckman, Hamilton, Tecan, Caliper, Perkin Elmer, Dynamic Devices, Agilent and Eppendorf) providing robustness and reproducibility. Plasmid yields may vary according to E.coli strain used, plasmid copy number and growth conditions. Typically an overnight culture in 1 mL LB medium provides 10 up to 15 pg of plasmid DNA for high-copy plasmids. The isolated plasmid DNA is endotoxin free and directly suitable for transfections, Sanger sequencing, restriction enzyme digestion as well as other molecular biology procedures. Bacteria are resuspended and then lysed using our CleanNA buffer system. Once the lysis process has been stopped, the samples are centrifuged and a cleared supernatant is transferred into a new processing plate. Optional, our lysate clearance beads can be used to clear the lysate from any cell debris. Plasmid DNA is then bound to our magnetic beads in presence of our binding buffer. The CleanNA particles are separated from solution using a magnetic device. After a few wash steps, the purified plasmid DNA is eluted from the beads using an elution buffer. • Sanger Sequencing • Restriction digestion Coenecoop 75 2741 PH Waddinxveen The Netherlands
