Instrumental Response and Measurement Capabilities
The document discusses the capabilities of different FS5 models, focusing on the instrumental response width, which can be enhanced with a faster detector (FS5-TCSPC+). Using an EPL as an excitation source, the response width can be approximately 250 ps. The document also highlights the ability to perform MCS measurements on microsecond and millisecond time scales, particularly in immune-assays where the FRET rate of donor-acceptor pairs depends on antigen concentration.
Measurement Examples
Examples include using an EPLED 280 with a standard detector for Norharmane in ethanol, and an EPL 405 with a faster detector for Hematoporphyrine in PBS.
Measurement Modules and Accessories
The FS5 supports various measurement modules and accessories, including cryostat cassettes, calibration kits, and cuvette holders. These modules are user-installable, and the software automatically recognizes the module in use, simplifying the user interface.
Optics and Specifications
The optics are all-reflective for wavelength-independent focus with high brightness. The source is a 150 W CW ozone-free Xenon arc lamp. The spectral coverage for excitation is 230 nm to 1000 nm, and for emission, it is 200 nm to 870 nm. The system includes fully automated filter wheels and offers a high signal-to-noise ratio.
Upgrade Specifications
Specifications for various upgrades include FS5-MCS, FS5-TCSPC, FS5-NIR, and FS5-POL. The lifetime range for different sources is specified, with the TCSPC+ version offering a range from less than 25 ps to over 10 ms.

Thorough calibration procedures are used to guarantee correct data representation. Spectral calibration is used to ensure wavelength accuracy and repeatability and photometric calibration is used to ensure presentation of true spectra without distortion of instrumental throughput functions. Traceable standards from NIST and BAM are used for calibration throughout the full wavelength coverage to verify calibration, in photons per unit bandwidth, of recorded spectra. The FS5 delivers data you can trust and utilise for high impact results in the research and analytical markets. NIST standards (top)...

Measurement Examples Emission Scans - with temperature dependence Rhodamine B, unlike other Rhodamine derivatives, has a chemical structure that is not entirely rigid. The diethylamino groups are interacting with the solvent and their mobility is therefore coupled to the solvent temperature. This causes the fluorescence intensity to have a strong dependence on the sample temperature. The example shows this effect, measured with the TE cooled sample holder. Measurements of absolute fluorescence quantum yield Fluorescence quantum yields can be measured by using the optional integrating sphere. The absolute method requires two measurements; the number of absorbed photons and the number of the emitted photons. The number of absorbed photons of a sample is determined by the reduction of the light scatter compared to a blank measurement. The quantum yield calculation is made using a wizard within the operating software. Quinine bisulphate in perchloric acid. The red curve shows the scan over the excitation scatter at 350 nm and the emission of the sample,...

Upgrade Options The upgrade options are modifications or additions to the main body of the FS5. All upgrades are best installed at the time of manufacture, but they can also be retro-fitted by a qualified service engineer during an on-site visit. All the standard features of the FS5 are retained when an upgrade option has been added. Many of the options can be combined. This offers outstanding flexibility for an instrument of this class.
