Methodology:
  • RPR VDRL Test: A serologic cardiolipin-based test for quick detection of Syphilis. It uses a suspension of cardiolipin/lecithin/cholesterol and carbon particles to visually detect antibodies (reagin) through the formation of black dumps.
  • TPHA Test: Detects T. pallidum antibodies using an haemagglutination method. Avian erythrocytes sensitized with T. pallidum fragments agglutinate in the presence of specific antibodies, forming a clouding effect in microtitre plates.
Procedures:
  • TPHA Qualitative Procedure: Mix 190 µL diluent with 10 µL serum, transfer 25 µL of mixture, add 75 µL of Test/Control Cells, and observe agglutination intensity.
  • RPR VDRL Qualitative Procedure: Dispense serum/plasma and antigen suspension, rotate for 8 minutes, and examine for agglutination.
Results Interpretation:
  • Negative results: No visible agglutination.
  • Positive results: Agglutination intensity from +/- to 4+.
  • All positive results should be confirmed with a semi-quantitative procedure.

Principle During Syphilitic infection, 2 groups of antibodies are detectable: nontreponemal antibodies which react with nonspecific antigens (RPR VDRL test), and treponemal antibodies which react with the specific antigens to T. pallidum (TPHA test). RPR VDRL test is a serologic cardiolopidic and nontreponemic test for the quick detection of Syphilis.
