Human identification High DNA Content Flexplex™ BioChipSet™ Cassette The High DNA Content Flexplex BioChipSet Cassette enables reference buccal samples to be processed using a 27-locus short tandem repeat (STR) assay with the DNAscan™ 6C Rapid DNA Analysis™ System from GE Healthcare and NetBio. The additional data from the larger Flexplex locus set offers greater discriminatory power and supports compatibility with DNA databases around the world. The system will process up to five samples and automatically generate an STR profile in less than 90 minutes using the on‑board Expert System software. The High DNA Content Flexplex BioChipSet Cassette offers the following features: • Expanded STR locus set: The Flexplex chemistry, optimized for rapid STR analysis, includes 27 loci for increased profile discrimination and compatibility with DNA databases around the world (Fig 1). • All-in-one consumable: The single-use NetBio BioChipSet Cassette contains all of the reagents necessary for STR DNA analysis, eliminating the need for loading or handling reagents, which minimizes contamination risk. Each cassette also contains an allelic ladder and internal lane standard. Fig 1. Configuration of the Flexplex STR chemistry. The Flexplex assay The High DNA Content Flexplex BioChipSet Cassette and DNAscan 6C Rapid DNA Analysis System have been developed to support compatibility with DNA databases around the world. As countries develop their DNA databasing requirements (e.g., the introduction of an Extended European Standard Set and the intended expansion of the core CODIS loci in the United States), STR chemistries must advance to meet the needs of Rapid DNA users. • Sample control: NetBio BioChipSet Swabs include an RFID tag for sample tracking and lock into the cassette sample chambers to minimize the potential for sample mix-ups. • Contamination control: All aspects of sample processing, including waste containment, are performed on the single-use cassette. The NetBio BioChipSet Cassette is a closed system with individual microfluidic channels for each sample, reducing contamination risk. • Room temperature storage: The preloaded reagents onboard the NetBio BioChipSet Cassette are room temperature stabilized, and therefore do not require refrigeration.

Flexplex STR chemistry supports these requirements by containing 23 autosomal loci, including SE33 and D6S1043, to maximize global DNA database coverage. Further inclusion of three Y-chromosomal loci (DYS391, DYS576, and DYS570) and Amelogenin provides both increased sample discrimination and improved compatibility across international databases.
