Standard Method Agar(SPC: Standard Plate Count (for viable count)) Code Directions Suspend 23.5 g of the dehydrated medium in 1,000 mL of distilled water, mix well and heat to dissolve the medium. Sterilize by autoclaving at 121 °C for 15 minutes. Maintain the medium at about 50 °C. Distribute about 20 mL into Petri dishes contained test samples in advance, and mix well. Incubate at 35 ± 1 °C for 48 ± 3 hours. Remarks The medium is applied for the following purposes; enumeration of bacteria in milk, dairy products, soft drink powder, chipped ice, ice cream, oysters, frozen fishes and seafood products and others.
