Introduction
The Pseudalert* test is a diagnostic tool for detecting Pseudomonas aeruginosa in water samples. It uses bacterial enzyme detection technology, which causes substrate hydrolysis and results in blue fluorescence under UV light, indicating the presence of the bacteria. The test is sensitive enough to detect as low as 1 cfu in 100 mL or 250 mL samples within 24 hours.

Storage
Test kits should be stored at temperatures between 2–30°C and kept away from light.

Presence/Absence (P/A) Procedure
1. Add the contents of a snap pack to a 100 mL or 250 mL water sample in a sterile, non-fluorescing vessel.
2. Cap and shake the vessel.
3. Incubate at 38±0.5°C for 24 to 28 hours.
4. Use the Result Interpretation table to interpret results.

Quanti-Tray* Enumeration Procedure (100 mL Samples Only)
1. Add the contents of a pack to a 100 mL water sample in a sterile vessel.
2. Cap and shake until dissolved.
3. Add 2 drops of IDEXX Antifoam Solution.
4. Pour the mixture into a Quanti-Tray* and seal.
5. Incubate at 38±0.5°C for 24 to 28 hours.
6. Count positive wells and refer to the MPN table for results.

Result Interpretation
- No blue fluorescence indicates a negative result for Pseudomonas aeruginosa.
- Blue fluorescence indicates a positive result for Pseudomonas aeruginosa.
A 6-watt, 365-nm UV light should be used for detection.

Procedural Notes
- Use sterile, non-buffered, oxidant-free water for dilutions.
- The test is not validated for flavored, marine, or carbonated water.
- Follow aseptic techniques and dispose of materials according to Good Laboratory Practices.

Quality Control Procedures
1. Perform quality control for each lot using IDEXX-QC Pseudomonas or additional methods.
2. Follow the P/A or Quanti-Tray Enumeration Procedure.
3. Results should align with the Result Interpretation table.
