ON-­‐LINE QUANTITATIVE CELL IMAGING ANALYSER TECHNOLOGY Ovizio’s platform is based on patented “Differential Digital Holographic Microscopy”. This unique technology brings real-­‐time label-­‐free monitoring to suspension cell bioreactors. In Digital Holography, the scattered light beam from an illuminated object interferes with a reference beam on a CCD camera allowing for a 3D numerical reconstruction of the object. Differential Digital Holography is an evolution of this base technology that brings increased stability and an important size reduction of the instruments. Digital Holography can construct intensity images; quantitative phase images and 3D phase images covering the shape and density of an object in a volume. The phase image is equivalent to the optical thickness of the sample. Images of cells captured in a hologram can be refocused after acquisition. The iLine F measures the specific holographic signature of each cell within the field of view and computes a cell count based on a sequence of holograms. The platform reports cell density and cell viability per measurement sequence (on-­‐line). Ovizio’s instrument is designed with bioreactors in mind. The technology can track a bioreactor run and counts cell viability from very low to high cell concentrations. This device has been developed for direct monitoring of suspension cell bioreactors. The device has built-­‐in functionality to monitor cell density and cell viability in real-­‐time. The iLine F can be connected to a bioreactor via an innovative disposable fluidics system, BioConnect. The advanced, easy to use software captures real time 3D images and quantitative data allowing for automatic cell counting and other specific measurements. The total magnification is typically 20 X but can be adapted to fit the size of the cells to be observed. Tracking cell growth continuously allows for more accurate prediction of the optimum harvest times. The platform is designed to cope with very low cell densities and very high densities. Quantitative imaging as a new Process Analytical Technology (PAT) is applied to on-­‐line bioreactor monitoring and improves process knowledge, quality and productivity in R&D, Pilot and Production.
