Mastocytosis is a group of diseases characterized by the abnormal accumulation of neoplastic mast cells in tissues and organs. The KIT D816V mutation is a common marker in mastocytosis patients and is part of the WHO diagnostic criteria.

PlentiPlex™ Mastocytosis Assay
The PlentiPlex™ Mastocytosis assay combines the selectivity of SensiScreen® FFPE assays with the sensitivity of SensiScreen® Liquid, allowing for the detection of single copy mutants in DNA samples. It can detect a single copy of mutated DNA in 100 ng of wild-type (WT) DNA with a limit of detection (LOD) of 0.003%. The workflow involves purifying DNA from a biopsy, adding it to ready-to-use tubes, running PCR, and analyzing the results using a MyGo Pro qPCR instrument.

Microsatellite Instability (MSI) Assays
Microsatellites are short nucleotide sequences prone to replication errors, leading to microsatellite instability (MSI) if not corrected by the DNA mismatch repair system. PlentiPlex™ MSI assays are used for diagnosing impaired DNA mismatch repair systems, affecting cancer prognosis and treatment selection. The assays offer three multiplexed MSI assays for analyzing five microsatellite loci, following Bethesda guidelines. The workflow includes purifying tumor and WT DNA, adding templates to reaction tubes, running PCR, and evaluating PCR product lengths on a DNA sequencing instrument.

PlentiPlex™ MYD88 L265P Assay
This assay is designed for diagnosing the leucine to proline mutation in codon 265 of the MYD88 protein in genomic DNA samples. It assists in distinguishing between Lymphoplasmacytic lymphoma/Waldenstrom macroglobulinemia (LPL/WM) and non-Hodgkin lymphoma. The assay combines high sensitivity with ease of use, working on standard real-time PCR equipment. It uses INA® technologies and BaseBlockers™ to suppress false positives. The workflow involves purifying gDNA, adding templates to reaction tubes, running PCR, and analyzing data on a MyGo Pro qPCR instrument.
