Technical Data Sheet Cl-Inhibitor Plus For In Vitro Diagnostic Use The MicroVue Cl-Inhibitor Plus EIA provides quantitative measurement of functional Cl-Inhibitor protein in experimental samples. C1 -Inhibitor is a multi-specific protease inhibitor present in human plasma and serum. Among the proteins it regulates are C1 and C1r of the C1 complemet complex. Genetic deficiencies of C1 -Inhibitor are of two types: functional deficiencies (protein is present but not functional) and antigen deficiencies (protein not present, or more commonly present at a low level). Either of these may lead to life threatening angioedema (HAE). Format ■ ELISA ■ 96-well assay plate ■ Sample type: Serum, EDTA plasma ■ Controls: Normal, abnormal included Species Reactivity ■ Rhesus Macaque, Baboon, Human ■ No reactivity with Cow, Chicken, Goat, Mouse, Rabbit, Rat, Sheep, Guinea Pig Specimen ■ Serum or EDTA plasma samples collected to avoid hemolysis Assay Steps ■ Dilute Wash Buffer; reconstitute Standards, Controls and C1 -Inhibitor Reactant ■ Dilute samples 1:101 ■ Pipette 100 pL of Standards, Controls and diluted samples into micro-tubes. ■ Add 20 pL C1 -Inhibitor Reactant into micro-tubes. Vigorously vortex. ■ Incubate 30 ± 1 minutes at 15°C to 30°C ■ Pipette 50 pL C1 -Inhibitor Reactant-treated Standards, Controls and specimen into microassay wells. ■ Incubate 10 ± 1 minutes at 15°C to 30°C ■ Wash the assay wells five times ■ Add 50 pL of C1 -Inhibitor Conjugate to each well ■ Incubate 60 ± 1 minutes at 15°C to 30°C ■ Wash the assay wells five times ■ Pipette 100 pL Substrate Solution to each assay well ■ Incubate 15 minutes at 15°C to 30°C ■ Add 100 pL of Stop Solution to each assay well ■ Measure absorbance at 450 nm Assay Performance Method: ELISA Analyte: Functional C1 -Inhibitor Protein Sample Volume: Minimum of 10 pL Abnormal Results: >40% Mean Normal Precision (inter-assay): 5.7%-10.0% Precision (intra-assay): 3.3%-5.4% Assay Time: Approx. 3 Hours Quidel | Specialty Products | Research to Rapids® | quidel.com | 800.874.1517 | 858.552.1100 MicroVue C1 Inhibitor Plus EIA - Cat. #A03
