Reagent Preparation
Bring all reagents to room temperature before use. Preparation instructions for specific reagents include:

Reagent Diluent Concentrate 2 (10X): Dilute 1:10 in deionized water for 1% BSA in PBS or 1:100 with Tris Buffer + 0.05% Tween® 20 for 0.1% BSA in TBS.
TMB ELISA Substrate: Protect from light.
1X Wash Buffer: Warm to room temperature if crystals form and mix gently. Prepare by adding 20 mL of concentrate to 480 mL of water.

Do not use past kit expiration date. * Provided this is within the expiration date of the kit. REAGENT PREPARATION Bring all reagents to room temperature before use. Note: Reagent preparations listed below are sufficient for one 96-well plate. Adjust volumes accordingly. Reagent Diluent Concentrate 2 (10X): • For assays that require 1% BSA in PBS, dilute 1:10 in deionized water. • For assays that require 0.1% BSA in TBS, dilute 1:100 with Tris Buffer + 0.05% Tween® 20, pH 7.2-7.4. Note: The Reagent Diluent will require additional components for some DuoSets. Refer to the product-specific package insert for more information. TMB ELISA Substrate - Protect from light. 1X Wash Buffer - If crystals have formed in the concentrate, warm to room temperature and mix gently until the crystals have gone into solution. Add 20 mL of Wash Buffer Concentrate to 480 mL of deionized or distilled water to prepare 500 mL of 1X Wash Buffer. PRECAUTIONS The Stop Solution provided with this kit is an acid solution. Some components in this kit contain a preservative which may cause an allergic skin reaction. Avoid breathing mist. Wear protective gloves, clothing, eye, and face protection.
