Human High Sensitivity Cytokine Base Kit A

Product catalog summary
Introduction
Cytokines are essential proteins for intercellular signaling, playing roles in growth regulation, cellular proliferation, and immune response modulation. The Human High Sensitivity Cytokine Base Kit A is designed for quantifying multiple human cytokines in serum and plasma using Luminex® technology.

Principle of the Assay
The assay uses analyte-specific antibodies on magnetic microparticles with fluorophores, analyzed by a Luminex system. Analytes bind to antibodies, followed by washes and a biotinylated antibody cocktail. Streptavidin-phycoerythrin is added, and dual laser analysis measures bound analytes.

Limitations of the Procedure
This research-only kit is unsuitable for diagnostics. It must be used before expiration, and reagents from different lots should not be mixed. Sample handling and assay conditions can affect results, and untested factors may interfere.

Materials Provided & Storage Conditions
The kit includes cytokine standards, diluents, and wash buffer, stored at 2-8°C. Reconstituted materials should be freshly prepared for each assay.

Other Supplies Required
Necessary equipment includes a Luminex analyzer, microplate magnet, pipettes, and a microplate shaker. Safety equipment is recommended.

Precautions
Some components contain preservatives that may cause allergies. Protective gear should be worn, and hands washed after handling.

Technical Hints
Avoid foaming when mixing, change pipette tips between additions, and protect microparticles from light for accurate results.

Sample Collection & Storage
Serum and plasma samples should be collected and stored properly to maintain stability, avoiding repeated freeze-thaw cycles.

Sample Preparation
Samples need centrifugation and dilution, with a 2-fold dilution recommended using Calibrator Diluent RD6-40.

Reagent Preparation
Reagents should reach room temperature before use. Wash buffer and standards must be prepared as instructed for proper assay performance.

Diluted Microparticle Cocktail Preparation
Microparticle concentrates should be centrifuged, vortexed, and diluted, used within 30 minutes, and protected from light.

Biotin-Antibody Concentrates Preparation
Biotin-Antibody Concentrates are prepared by centrifuging, vortexing, and diluting in Biotin Antibody Diluent 2, with specific volumes for different well counts.

Instrument Settings
Instructions for MAGPIX® and Luminex® analyzers include sample volume, microparticle region assignment, and parameters like flow rate and reporter gain.

Assay Procedure
Reagents and samples are prepared, added to wells, and incubated with a microparticle cocktail. The process includes washing steps, adding Biotin-Antibody Cocktail and Streptavidin-PE, followed by incubation and analysis. Light protection for certain components is crucial.

Calculation of Results
Results are calculated using standard concentrations and a standard curve with a five-parameter logistic curve-fit. Concentrations must be adjusted by the dilution factor.

Calibration
The assay is calibrated against recombinant human cytokines from R&D Systems®.

Specificity
The assay shows less than 0.5% interference with various recombinant proteins.

Plate Layout
A plate layout is provided for recording standards and samples.

Notes
The assay is for research use only, not for diagnostic procedures, with trademark information included.

INTRODUCTION Cytokines are intercellular signaling proteins released from a wide variety of cells and tissues. They play an integral role in regulating growth and cellular proliferation as well as modulating host response to infection, injury, and inflammation. Cytokines also influence reproduction and bone remodeling. A large number of cytokines are pleiotropic and share similar functions. In addition, many cytokines influence the production of other cytokines. Analysis and quantification of cytokines in biological fluids has thus become increasingly important. Methods such as bioassay, enzyme-linked...

PRINCIPLE OF THE ASSAY Luminex® Performance Assay multiplex kits are designed for use with any Luminex analyzer including the MAGPIX®, Luminex® 100/200™, FLEXMAP 3D®, xMAP INTELLIFLEX®, or Bio-Rad® Bio-Plex®, dual laser, flow-based sorting and detection platforms. Analyte-specific antibodies are pre-coated onto magnetic microparticles embedded with fluorophores at set ratios for each unique microparticle region. Microparticles, standards and samples are pipetted into wells and the immobilized antibodies bind the analytes of interest. After washing away any unbound substances, a biotinylated antibody...
