Luminex® Performance Assay - Human Cytokine Base Kit A

Product catalog summary
Introduction
This document describes a cytokine assay kit designed for the simultaneous quantification of multiple cytokines in biological samples. It addresses the limitations of traditional methods like ELISA and PCR, which cannot measure multiple cytokines at once.
Principle of the Assay
The assay utilizes Luminex® technology with magnetic microparticles coated with analyte-specific antibodies. These are analyzed using lasers to identify microparticle regions and quantify analytes. The process involves binding analytes, adding a biotinylated antibody cocktail, and detecting with streptavidin-phycoerythrin.
Limitations of the Procedure
The kit is intended for research use only and not for diagnostic purposes. It should not be used after expiration, and reagents from different lots should not be mixed. Variations in sample handling can affect results, and while designed to minimize interference, this cannot be guaranteed without testing.
Materials Provided & Storage Conditions
The kit includes reagents like standard cocktails and wash buffer, which should be stored at 2-8°C and used before expiration. Fresh solutions are required for each assay.
Other Supplies Required
Necessary equipment includes a Luminex analyzer, microplate magnet, pipettes, and a microplate shaker. Optional controls are available.
Precautions
Some components contain preservatives that may cause allergic reactions. Protective gear is recommended, and hands should be washed after handling.
Technical Hints
Avoid foaming when mixing, change pipette tips to prevent contamination, and protect reagents from light.
Sample Collection & Storage
Guidelines are provided for collecting and storing samples like cell culture supernates, serum, and plasma, which should be stored at ≤ -20°C to avoid freeze-thaw cycles.
Sample Preparation
Serum and plasma samples require centrifugation and dilution, with specific instructions for different analytes.
Reagent Preparation
Reagents should reach room temperature before use. Instructions are given for preparing wash buffer and calibrator diluents, with standards reconstituted as per value cards.
Standard Preparation
A 3-fold dilution series is prepared using reconstituted Standard Cocktail vials, with working standard 1 as the high standard and a calibrator diluent as the blank.
Microparticle Cocktail Preparation
Microparticle Concentrates are centrifuged, vortexed, and diluted, with specific volumes for different well numbers. Protection from light is crucial.
Biotin-Antibody Cocktail Preparation
Similar preparation steps as microparticles, with specified volumes based on well numbers.
Streptavidin-PE Preparation
Involves centrifuging, vortexing, and diluting in Wash Buffer, with specific volumes for different well numbers, emphasizing light protection.
Instrument Settings
Detailed settings for analyzers are provided, including sample volume, bead type, and calibration requirements.
Assay Procedure
The procedure includes reagent preparation, sample addition, incubation, washing, and reading results with analyzers, stressing light protection.
Calculation of Results
Results are calculated using standard concentrations and a 5-parameter logistic curve-fit, considering dilution factors.
Specificity
The assay shows less than 0.5% cross-reactivity with various recombinant proteins.
Plate Layout
A layout is provided for recording standards and samples.

Luminex® Performance Assay - Human Cytokine Base Kit A-3

INTRODUCTION Cytokines are intercellular signaling proteins released from a wide variety of cells and tissues. They play an integral role in regulating growth and cellular proliferation as well as modulating host response to infection, injury, and inflammation. Cytokines also influence reproduction and bone remodeling. A large number of cytokines are pleiotropic and share similar functions. In addition, many cytokines influence the production of other cytokines. Analysis and quantification of cytokines in biological fluids and cell culture supernates has thus become increasingly important. Methods...

PRINCIPLE OF THE ASSAY Luminex® Performance Assay multiplex kits are designed for use with any Luminex analyzer including the MAGPIX®, Luminex® 100/200™, FLEXMAP 3D®, xMAP INTELLIFLEX®, or Bio-Rad® Bio-Plex®, dual laser, flow-based sorting and detection platforms. Analyte-specific antibodies are pre-coated onto magnetic microparticles embedded with fluorophores at set ratios for each unique microparticle region. Microparticles, standards and samples are pipetted into wells and the immobilized antibodies bind the analytes of interest. After washing away any unbound substances, a biotinylated antibody...
