Product catalog summary
Specifications
The document describes the specifications of a CHO-derived human IL-23 protein. The protein consists of two subunits: p40 and p19, with accession numbers P29460.1 and AAG37232.1, respectively. The predicted molecular masses are 55 kDa for the IL-23 single chain, 37 kDa for p40, and 19 kDa for p19. SDS-PAGE analysis shows molecular weights of 59-68 kDa, 46-52 kDa, and 28-33 kDa under reducing conditions. The protein's activity is measured by its ability to induce IL-17 secretion by mouse splenocytes, with an ED50 of 0.100-1.00 ng/mL. The endotoxin level is less than 0.10 EU per 1 μg of protein, and purity is greater than 95% as determined by SDS-PAGE and staining methods.
Preparation and Storage
The protein is lyophilized from a filtered solution in PBS with Trehalose. For reconstitution, a 10 μg size should be reconstituted at 100 μg/mL in PBS, while other sizes should be reconstituted at 500 μg/mL. The product is shipped at ambient temperature and should be stored at -20 to -70 °C for up to 12 months. After reconstitution, it can be stored for 1 month at 2 to 8 °C or for 3 months at -20 to -70 °C under sterile conditions.
Data
The molecular weight of the recombinant human IL-23 protein, as analyzed by SEC-MALS, is 57.7 kDa, indicating it is a heterodimer. This may differ from the predicted molecular weight due to post-translational modifications such as glycosylation. The bioactivity of the protein is confirmed by its ability to induce IL-17 secretion by mouse splenocytes, with an ED50 of 0.100-1.00 ng/mL.
Background
IL-23 is a heterodimeric cytokine composed of a unique p19 subunit and a p40 subunit shared with IL-12. The p19 subunit is homologous to the p35 subunit of IL-12 and other cytokines like IL-6 and IL-11. The p40 subunit is similar to the extracellular domains of hematopoietic cytokine receptors. IL-23 is produced by activated macrophages and dendritic cells and acts on memory T cells to induce IL-17 secretion, a proinflammatory cytokine. Unlike IL-12, IL-23 specifically acts on CD8+ dendritic cells to mediate immunogenic presentation of poorly immunogenic peptides.

Reconstitute all the other sizes at 500 pg/mL in PBS. Shipping The product is shipped at ambient temperature. Upon receipt, store it immediately at the temperature recommended below. Stability & Storage Use a manual defrost freezer and avoid repeated freeze-thaw cycles. • 12 months from date of receipt, -20 to -70 °C as supplied. • 1 month, 2 to 8 °C under sterile conditions after reconstitution. • 3 months, -20 to -70 °C under sterile conditions after reconstitution. Recombinant Human IL-23 (CHO Expressed) Protein SEC-MALS. Recombinant Human IL-23 (Catalog # 11349-IL) has a molecular weight (MW) of 57.7 kDa as analyzed by SEC-MALS, suggesting that this protein is a heterodimer. MW may differ from predicted MW due to post-translational modifications (PTMs) present (i.e. Glycosylation).

Recombinant Human IL‑23 (CHO Expressed)-2

Recombinant Human IL-23 (CHO Expressed) Catalog Number: 11349-IL BACKGROUND Interleukin 23 (IL-23) is a heterodimeric cytokine composed of two disulfide-linked subunits, a p19 subunit that is unique to IL-23, and a p40 subunit that is shared with IL-12 (1-5). The p19 subunit has homology to the p35 subunit of IL-12, as well as to other single chain cytokines such as IL-6 and IL-11. The p40 subunit is homologous to the extracellular domains of the hematopoietic cytokine receptors. Human p19 cDNA encodes a 189 amino acid residue (aa) precursor protein with a putative 19 aa signal peptide and 170...
