Principle and Interpretation: Cetrimide, a quaternary ammonium compound, exhibits bactericidal activity against many Gram-positive and some Gram-negative organisms. Pseudomonas aeruginosa produces iron chelators like pyoverdin, which, when combined with pyocyanin, results in a characteristic bright green color. Magnesium chloride and potassium sulphate enhance chelator production.
Composition: The medium includes gelatin peptone, magnesium chloride, potassium sulphate, cetrimide, and agar, with a pH of 7.2 ± 0.2.
Precautions: For professional use only. Do not use if plates show contamination or deterioration.
Test Procedure: Inoculate plates using streak or spread method and incubate at 35-37°C for up to 48 hours. Confirm presumptive identification with additional tests.
Quality Control:
  • Sterility Control: No growth after incubation at specified conditions.
  • Physical/Chemical Control: Light amber appearance with a pH of 7.2 ± 0.2.
  • Microbiological Control: Good growth of Pseudomonas aeruginosa, partial inhibition of E. coli, and inhibition of S. aureus.
Limitations: The medium is for primary isolation. Pure cultures are recommended for further biochemical and serological tests.
Storage and Shelf Life: Store at 2-12°C and use before the expiry date.
Disposal: Infected media should be autoclaved, incinerated, or treated with a chlorine-based disinfectant before disposal.

PRINCIPLE AND INTERPRETATION: Cetrimide is a quarternary ammonium compound with bactericidal activity against a broad range of Gram-positive organisms and some Gram-negative organisms. Pseudomonas aeruginosa produces a number of water soluble iron chelators, including the yellow-green or yellow-brown fluorescent pyoverdin. When pyoverdin combines with the blue water-soluble pyocyanin, the bright green colour characteristic of Pseudomonas aeruginosa is created. The addition of magnesium chloride and potassium sulphate enhances the production of these chelators. COMPOSITION: LIMITATIONS OF THE PROCEDURE: This prepared plated medium is intended for primary isolation. Some diagnostic tests may be performed with the primary plate. The agents in selective media may inhibit some strains of the desired species or permit growth of a species they were designed to inhibit, especially if the species is present in large numbers in the specimen.
