Technical Note High Density Microfluidic SCOPE-chip™ Boost cell throughput on one chip This technical note describes Singleron’s High Density (HD) Microfluidic SCOPE-chip™, a microwell chip for single-cell dispersion, mRNA capture, and barcoding. Its performance is evaluated focusing on coverage of all cell types, consistency, and doublet rates. Overview High-throughput single-cell sequencing technologies have advanced rapidly during the past decade. Technical Note High Consistency To evaluate the consistency of data, when processing samples on the HD chip, three mouse testis samples were processed and analyzed following the standard GEXSCOPE workflow, simultaneously. At a sequencing depth of 13,200 reads/cell, the average number of cells detected was 21,289 cells, the mean gene number was 1350 genes, and the average number of total genes was 31,116 genes. Technical Note Capture of Rare Cell Types To investigate the ability of Singleron’s HD SCOPEchip to capture rare cell types, mouse embryonic cells were processed on two HD SCOPE-chips, then libraries were pooled and analyzed at a sequencing depth of 21,900 reads per cell. A total of 42,628 cells was detected with a median of 1677 genes per cell and 31,917 genes, cumulatively. Technical Note Low Doublet Rate In order to evaluate occurring doublet rates, Singleron’s HD SCOPE-chip technology was compared to another commercially available platform (Figure 4). While higher numbers of captured cells come with an enormous increase in data output, doublet rates only moderately rise. Thus, Singleron’s HD SCOPE-chip technology outperformed the other technology.
